Myelo Monocytic Cell Marker : CD 14

CD14 is a cell-surface pattern-recognition receptor and a marker for monocytes and macrophages (and to a lesser extent neutrophils and some dendritic cells). Present as membrane-bound and soluble forms, it helps recognize bacterial LPS with TLR4/MD-2 and is used clinically and in research to identify monocyte-lineage cells and indicate innate immune activation or inflammation.

Yes. Human monocytes constitutively express CD44 (the hyaluronan receptor) on their surface; expression commonly increases with activation and during differentiation into macrophages. CD44 mediates adhesion, migration, and signaling in inflammatory responses. Alternative splice isoforms and regulated up‑regulation occur in activated or tissue‑resident macrophages, supporting immune-cell trafficking and tissue remodeling.

CD14-positive monocytes are circulating white blood cells that express the CD14 receptor, a pattern-recognition co-receptor for bacterial lipopolysaccharide. They play key roles in innate immunity by sensing pathogens, producing inflammatory cytokines (e.g., TNF-α, IL-1, IL-6), and differentiating into macrophages or dendritic cells. Subsets include classical (CD14++CD16−), intermediate (CD14++CD16+), and non-classical (CD14+CD16++).

Monocytes typically express CD14 and CD16, defining classical (CD14++CD16−), intermediate (CD14++CD16+), and nonclassical (CD14+CD16++) subsets. Other common markers include CD64 (FcγRI), CD11b/CD18 integrins, HLA-DR (MHC II), CD36, CCR2 (chemokine receptor), and CSF1R (CD115). Marker expression varies with activation state and tissue recruitment. These surface proteins are used clinically and in research to identify, classify and track monocyte subsets in blood and inflamed tissues.

Common CD markers for myeloid-lineage cells include CD13 and CD33 (myeloid progenitors and mature cells), CD11b and CD11c (integrins on monocytes, macrophages, granulocytes, dendritic cells), CD14 and CD64 (monocytes/macrophages), CD15 and CD66b (granulocytes), CD16 (neutrophils/monocyte subsets), and CD117/CD34 on progenitors. These markers are used in flow cytometry to define lineage and maturation.

CD14 and CD16 mark monocyte subsets: classical CD14++CD16− monocytes (~80–90%) are highly phagocytic, CCR2-driven, and recruited rapidly to inflamed tissues for acute responses. CD16+ monocytes (CD14+CD16+ intermediate and CD14dimCD16++ nonclassical) patrol endothelium, have lower phagocytosis but greater antigen-presentation/cytokine and antiviral responses, and contribute to vascular surveillance and chronic inflammation.
