CD3 (NATURAL KILLER CELLS)

Natural killer (NK) cells are typically CD3‑negative; CD3 is a T‑cell receptor complex not expressed on conventional NK cells. Human NK cells commonly express CD56 and CD16 instead. However, a distinct population called NKT cells co‑express T‑cell markers (including CD3) and NK markers — these are immunologically different from classic CD3‑negative NK cells.

Natural killer (NK) cells do not express CD3; they are CD3-negative innate lymphocytes. Human NK cells are typically CD56+ and may express CD16, but they lack the T-cell receptor–CD3 complex used by T lymphocytes. A distinct population, invariant NKT cells, co-express T-cell markers including CD3 and NK markers, but conventional NK cells remain CD3−.

Natural killer (NK) cells are a type of large granular lymphocyte of the innate immune system that develop in bone marrow. They kill virus-infected and cancerous cells without prior sensitization by recognizing stressed cells or missing MHC I. NK cells use perforin and granzymes to induce target-cell death and secrete cytokines like interferon-gamma to modulate immune responses.

CD3+ CD56+ cells are T lymphocytes that also express the NK-cell marker CD56 (often called NKT-like or NK-like T cells). They combine T-cell antigen recognition with innate-like cytotoxicity and rapid cytokine release, bridging adaptive and innate immunity. Present in blood and tissues, they participate in antiviral and antitumor responses and can be altered in infections, cancer, and autoimmune conditions.

If CD3 is positive, it indicates the presence of T lymphocytes—CD3 is a pan–T‑cell marker. This often reflects normal or reactive T‑cell populations; however, unusual patterns or aberrant co‑expression with other markers may suggest T‑cell malignancy. Results must be interpreted with CD4/CD8/CD20 and clinical context; further clonality/genetic testing and hematology referral may be needed.

The primary marker used to identify natural killer (NK) cells in tumors is CD56 (NCAM), typically expressed alongside absence of CD3 (CD56+ CD3−). Many NK cells also express CD16 (FcγRIII). Additional NK lineage/activation markers include NKp46 (NCR1), NKG2D and CD57, which help distinguish subsets and activation state within the tumor microenvironment. These markers guide immunophenotyping and research.
